Slabý, Tomáš ; Kršek, Jiří (referee) ; Chmelík, Radim (advisor)
The diploma thesis deals with a design of illuminating system for tandem-scanning confocal microscope using a high-power LEDs.
Ferrous nanoparts localization in inner organelles
Solař, Jan ; Janoušek, Oto (referee) ; Čmiel, Vratislav (advisor)
This bachelor‘s thesis describes a behavior of iron nanoparticles in human mesenchymal stem cells. First section deals with methods of fluorescent labelling of organelles and nanoparticles and settings of confocal microscope for their detection. Next section describes the iron nanoparticles metabolism and accumulation in cells. Finally, there is a section about the developement of the software utility for the localization and for the quantitative analysis of the fluorescence organelles and nanoparticles inside the living cells.
Utilization of confocal microscopy to study of degradation of organic dye in cell biology
Trnová, Kateřina ; Provazník, Ivo (referee) ; Čmiel, Vratislav (advisor)
This thesis focuses on the study of the life of a fluorescent dye. It also examines changes in its emission spectrum determined with the aid of development of fluorescence intensity over the long term sensing in several kinds if of cella. The general part of the thesis with the issues of confocal microscopy, fluorescent radiation and maks. Furthermorer, an analysis of methods for measuring fluorescence lifetime is done. In the thesis there is elaborated a description of the program, whitch was designed for the analysis of the collected output of the used confocal microscope. Sebsequently, the results are evaluated.
Classification of marked cells migration in tissue
Solař, Jan ; Skopalík, Josef (referee) ; Čmiel, Vratislav (advisor)
This diploma thesis deals with analysing of modern methods for cell detection, visualization and quantification in 3D space. The first section deals with optical methods for cells detection. There is detailed discussion about cell labeling and detection on confocal microscopy. There is also description about developed algorithm for whole cell volume quantification from microscopy images. This could made a comparsion of fluorescence signal according to time of cell labeling and according to cell shapes. There was also optimalization of handmade tissue phantoms visualization. It could be compared a possibilities of cell detections in these phantoms by confocal microscopy and OCT. It was also implemented algorithm for quantification of cells from OCT images. Besides confocal microscopy and OCT cells are also analyzed by other methods. The last part is the Conclusion of results and comparison of used methods.
System for measurement of concentration of intracellular calcium in LabVIEW
Pochlopeň, Timotej ; Ronzhina, Marina (referee) ; Čmiel, Vratislav (advisor)
Master‘s thesis deals with the methods and system for measurement of concentration of intracellular calcium in LabVIEW. The thesis covers the theory of fluorescence principles, fluorescence methods and indicators for measurement of concentration of intracellular calcium. The procedure of measurement by means of fluorescence microscope, camera and indicators is also mentioned. The theory dealing with confocal microscopy is presented and two techniques are compared. Literary solution is presented. The practical part describes the program with a graphical user interface used for acquisition and analysis of data measuring dynamics of intracellular calcium concentration. The experimental part describes the preparation and measurement of cells and evaluation of experiments.
Design of microscope LED illumination system
Gricová, Monika ; Dostál, Zbyněk (referee) ; Antoš, Martin (advisor)
The objective of this bachelor thesis was to design illumination assemblage in confocal microscope Confocal 2002. First part of this paper describes types and components of confocal microscopes. For correct function of the microscope, high-power illumination by a white light emmiter is required. Based on the analysis of the background research, suitable LED diode was selected. In experimental part of this work, eight optical systems are proposed, that has been assembled and evaluated. Based on the evaluation of obtained paramteres, the optimal assemblage was selected and designed for production.
Numerical Methods of Image Processing for Object Surface Reconstruction by Means of Confocal Microscope
Adámková, Barbora ; Martišek, Dalibor (referee) ; Štarha, Pavel (advisor)
The Diploma thesis deals with object surface reconstruction by means of confocal microscope. It includes part of mathematical theory which is associated with this problem. The mathematical algorithm of the object surface reconstruction is illustrated. This Diploma thesis result is the application development for this reconstruction. The thesis also include the results of specific object.
Evaluation of antibiotic-induced mitochondrial superoxide production in adherent cells
Ingrová, Kateřina ; Chmelíková, Larisa (referee) ; Zumberg, Inna (advisor)
The theoretical part of this bachelor´s thesis contains a description of the effect of reactive oxygen species on oxidative stress by mitochondria and the consequences of antibiotics use in cell line culturing. The cell line studied in this bachelor´s thesis is the mesenchymal stem cells (MSCs). The practical part describes the procedure of the experiment including cell culturing, passaging and cell labeling. The proposed experiment was repeated with sufficient number of repetitions. Finally, confocal microscopy images were processed in the MATLAB programming environment.
